TROPHOBLAST RETRIEVAL AND ISOLATION FROM THE CERVIX (TRIC) FORNON-INVASIVE PRENATAL GENETIC DIAGNOSIS AND PREDICTION OFABNORMAL PREGNANCY OUTCOMEby RANI B. FRITZAugust 2015Advisor: D. Randall Armant, Ph.D.Major: Physiology (Reproductive Sciences Concentration)Degree: Doctor of PhilosophyThe placenta is vital for the shortand long-term health of the fetus, and significantlyimpacts the health of the mother. During the first and second trimesters of pregnancy, extravillous trophoblast (EVT) cells invade the uterus and remodel the maternal spiral arterieswhich, if inadequate, lead to pregnancy complications, including early pregnancy loss (EPL),preeclampsia (PE), and intra-uterine growth restriction (IUGR). EVT migration into the uterinewall is dependent on growth factors and cytokines that signal between maternal and fetal tissues. The epidermal growth factor (EGF) signaling system plays a significant role in trophoblastsurvival and function. Using immunocytochemistry (ICC), we evaluated EGF family growth factors in post-partum PE placentas (villi and basal plate), and maternal serum, comparing toplacentas to gestational age (GA)-matched preterm pregnancies, IUGR pregnancies, and uncomplicated term pregnancies. EGF, heparin-binding EGF-like growth factor (HBEGF), and transforming growth factor alpha (TGFA) were significantly decreased in placentas from PEcompared to IUGR, preterm labor, and uncomplicated term placentas. It was uncertain whetherreduced growth factor signaling contributed to disease, or was the result of late-stage pathology. However, EGF was significantly reduced in prenatal serum of pregnant patients with PE

O-45: Quantification of Cell-Free-Fetal-DNAfrom Maternal Plasma for the First Time in Pakistan:Implications for Non-Invasive PrenatalDiagnosis of Genetic Disorders
Background: Current prenatal diagnosis requires invasive testing which carries a 1-4% procedure-related-risk of miscarriage; hence, non-invasive techniques are desired. The recent demonstration of cell-free-fetal-DNA enriched from maternal plasma has opened new possibilities for non-invasive-prenatal-diagnosis of not only genetic-disorders such as β-thalassaemia and haemophilia but also chromos...
متن کاملI-40: Non Invasive Prenatal Genetic Diagnosis;Current Status and The Future
Discovery of cell free fetal DNA in 1997 has deeply changed the outlook of prenatal diagnosis approaches as most of the clinically established screening tests are not sensitive/specific enough while the current practical diagnostic tests are also invasive in their nature. The most common prenatal screening test is routinely practiced for the diagnosis of Down syndrome (DS) which includes a 10% ...
متن کاملAltered Biomarkers in Trophoblast Cells Obtained Noninvasively Prior to Clinical Manifestation of Perinatal Disease
A contributing factor to poor placental perfusion, leading to intrauterine growth restriction and preeclampsia, is the failure of invading extravillous trophoblast (EVT) cells to remodel the maternal uterine arteries during the first and second trimesters of pregnancy. Noninvasive assessment of EVT cells in ongoing pregnancies is possible beginning three weeks after conception, using trophoblas...
